Abstract

The role of cortisol in different endocrine related diseases is quite established, which lead to the development of various selective 11ß-HSD1 inhibitors. To date, none of them has made it through clinical trials, mainly because of a lack of efficacy in the targeted disease. Recently, the intracrine role of cortisol in immune cells began to be investigated, opening a pathway for repurposing those drug candidates. The following work presents the synthesis of 11ß-HSD1 inhibitors and the development of their corresponding PROTAC. The compounds were selected among a list of different documented inhibitors to be engaged in a project whose aim is to provide a proof of concept that would demonstrate the role of 11ß-HSD1 in the immune system response to cancer.
