Measurement of Partition-Coefficient Using Centrifugal Partition Chromatography - Method Development and Application to the Determination of Solute Properties

Abstract

Measurement of partition coefficients in various solvent systems has been developed using flow-through centrifugal partition chromatography (CPC). The present paper details the experimental design, and shows that this method is able to measure compounds in a log P range of ca. -3.2 to 3.2 in a precise, accurate and rapid way. The usefulness of some lipophilicity-derived parameters such as Delta log P-oct-alk (log P-octanol minus log P-alkane, reflecting mainly the hydrogen bond donor acidity of solutes) was thus reinforced due to the timely development of this method. We also demonstrated the application of the Delta log P parameter both in characterizing the structural properties of drugs and in uncovering their pharmacological implications
