Here we show that the cytoplasmic junctional proteins cingulin (CGN) and paracingulin (CGNL1) interact directly with NM2s through their C-terminal coiled-coil sequences. CGN binds strongly to NM2B, and CGNL1 to NM2A and NM2B. Knockout (KO), exogenous expression, and rescue experiments with WT and mutant proteins show that the NM2-binding region of CGN is required for the junctional accumulation of NM2B, ZO-1, ZO-3, and phalloidin-labeled actin filaments, and for the maintenance of tight junction membrane tortuosity and apical membrane stiffness. CGNL1 expression promotes the junctional accumulation of both NM2A and NM2B and its KO results in myosin-dependent fragmentation of adherens junction complexes. These results reveal a mechanism for the junctional localization of NM2A and NM2B and indicate that, by binding to NM2s, CGN and CGNL1 mechanically couple the actomyosin cytoskeleton to junctional protein complexes to mechanoregulate the plasma membrane.</p> Dewey 616.07 Dewey 570 Swiss National Science Foundation Signaling at epithelial apical junctions 152899 CH Projects Swiss National Science Foundation Signalling by zonular junctional proteins in epithelia and endothelia 172809 CH Projects 7daed2ee-1311-4824-9df9-2e51260a966a Département de biologie moléculaire et cellulaire 16484 137 Département de pathologie et immunologie 21F false 1441 Groupe Citi 140 Groupe Christine Chaponnier (pathologie et immunologie) 171 The Journal of cell biology 222 7 d0d9cb55-bcfe-4839-8886-2ef299e99ce7 Article (Published version) Public Article-scientifique-from-crossref.pdf 10685177 application/pdf CC BY-4.0 d441fc7b53324ee93c98cdb192f5ae0ea3bfcf1cf062e9e9e78433aa5b7cfb96 openAccess Faculté des sciences / Section de biologie / Département de biologie moléculaire et cellulaire Faculté de médecine / Section de médecine fondamentale / Département de pathologie et immunologie
