Role of panx1 in bone marrow-derived macrophages and possible links to atherosclerosis

Abstract

Aim: Atherosclerosis is a major cardiovascular disease and the principal cause of death worldwide. It is a progressive disease of the medium-sized and large arties characterized by a dysfunctional endothelium, an accumulation of lipids and an abnormal inflammatory response. Extracellular ATP can act as a ‘find-me' signal to recruit macrophages at the site of inflammation. ATP can be released through Pannexin1 (Panx1) channels in a controlled manner. Panx1 channels have been linked to many inflammatory and vascular pathologies including thrombosis. However, the role of Panx1 in macrophages in the context of atherosclerosis remains unknown. Conclusion: Based on previous results obtained in the laboratory Panx1 seems to play a protective role in macrophages and/or endothelial cells during atherosclerotic plaque development. However, the present work highlights that Panx1 atheroprotective effects seem not due to changes in the adhesive properties of macrophages lacking Panx1.
