Abstract

The role of the DNA sequence on the nucleosome dynamics remains poorly understood, therefore t h e main biological question tackled by this project is: Does the DNA sequence contribute to nucleosome stability? To answer this question, in the following work we will generate a pool of plasmids with at least one million diferent yeast ribosomal protein gene RPS11A promoter variants, each one with altered, random 145 bp DNA sequence underneath the -1 FN, immediately upstream of an fuorescence reporter ORF. This library will then be transformed into budding yeast and the transformants will be pooled and sorted out according to the transcriptional output by fow cytometry. The DNA sequences underneath the -1 FNs will then be amplifed, subject to pairedend sequencing, and screened for motifs correlated with expression.
