Exploring the Role of MYB Transcription Factors in Endodermal Suberization in Arabidopsis thaliana

Abstract

Suberin is a hydrophobic biopolymer present around endodermal cells protecting the plant vasculature against stresses and controlling water and nutrient acquisition. Endodermal suberization is highly plastic, being regulated by environmental and developmental cues. Although suberin plasticity appears to be a hallmark of endodermal suberization, the molecular players involved remain largely unknown. In this context, we used endodermal-specific TRAP (Translating Ribosome Affinity Purification) to identify genes co-regulated with known suberin genes. Amongst these, we identified several additional MYBs, previously uncharacterized in the context of endodermal suberization. These candidates were validated through promoter reporter analysis and functional characterization. From within this selection, 4 MYBs were characterized in detail, comprehending 2 new positive regulators and 2 MYBs thought to be negative regulators of endodermal suberization.This thesis provides new information regarding the regulatory network of endodermal suberin, giving insights into its complex arrangement both in terms of transcription factors involved and necessary specific spatiotemporal dynamics. Moreover, the newly identified suberin-related MYBs could be placed in this network. Although far from untangling the network, this work encompasses different tools and observations crucial for the full understanding of endodermal suberin regulation.
