Abstract

Follicle-stimulating hormone (FSH) and its receptor (FSHR) are essential for ovarian folliculogenesis, but the role of FSHR internalisation and trafficking in downstream signalling remains unclear. This project investigates FSHR trafficking, hypothesising that FSHR routing through distinct intracellular compartments modulates downstream signalling pathways. Using FLAG-tagged mouse FSHR in immortalised mouse ovarian granulosa cells, we analysed receptor trafficking via immunofluorescence (colocalisation with intracellular compartment markers including APPL1 and EEA1) and examined FSH-induced activation of signalling pathways (MAPK, CREB and AKT) through Western blot analysis. FSHR was found to traffic through APPL1-positive and EEA1-positive endosomes during activation, with distinct signalling dynamics: rapid ERK1/2 phosphorylation, delayed CREB activation and sustained AKT signalling. These findings lay the foundation for further exploration of the role of FSHR trafficking in signalling.
