Abstract

Iontophoresis, a non-invasive technique, employs a low-intensity electric current to enable the controlled delivery of hydrosoluble charged molecules into and through the skin. Its effectiveness in enhancing drug delivery stems from the superior electrotransport of molecules compared to passive diffusion, achieved through electromigration or electroosmosis. Previous studies have demonstrated the successful delivery of moderately-sized cationic and anionic proteins, including antibodies, using this technology. However, relying solely on basic factors like molecular weight, net charge, or electric mobility is insufficient to predict protein delivery. The charge distribution on the surface of proteins, a more intricate parameter, could significantly influence protein electrotransport. This thesis aimed to explore the impact of charge distribution on protein surfaces on iontophoretic delivery. The investigation sought to enhance our understanding of the critical factors governing protein electrotransport, with the ultimate goal of identifying potential biotherapeutic candidates for iontophoresis.
