Development of sphingolipid probes for lipid dynamic observation during phagocytosis

Abstract

Sphingolipids have long been identified as bioactive lipids thus participating in cell regulation and signaling. Furthermore, several studies involved sphingolipids, notably ceramides and ceramide-1-phosphate in the process of phagocytosis. Among which a proteomic screen by Prof. Paula Nunes-Hasler (unpublished) showed enrichment of sphingolipid metabolism protein on STIM1 bearing phagosomes. Although evidence points toward the involvement of these lipids during phagocytosis, their exact function is yet to be established. Phagocytosis being an important process for both innate and acquired immunity, deciphering its mechanisms could lead to important strides regarding treatment of several diseases. Unfortunately, reliable tools such as fluorescent lipids probes to study sphingolipid dynamics in cells are lacking. Faced with this limitation and with the previous discovery highlighted by the proteomic screen, Prof. Nunes-Hasler's group decided to develop putative ceramide and ceramide-1-phosphate probes. The aim of this work was to characterize these probes and determine their potential as tools to study sphingolipid dynamics during phagocytosis. C-KSR and NES-PLA2G4-EGFP, respectively a ceramide probe and a ceramide-1-phosphate probe, were studied in the context of phagocytosis. NES-PLA2G4-EGFP did not arise as a strong ceramide-1-phosphate detector. However, C-KSR, though later found to be degraded, still emerged as a potential reliable ceramide detector. Nevertheless, modification will probably be required on the C-KSR construct to be used as such.
