Bioenergetic and metabolic profile of human muscle stem cells

Abstract

Satellite cells are endogenous Pax7+ muscle stem cells (MuSC), localized between the basal lamina and the sarcolemma of myofibers. MuSC are essential for supporting muscle regeneration after injury and represent a possible source for cell therapy in the treatment of myopathies. However, their amplification prior injection is known to reduce their regenerative potential. In the laboratory, we have demonstrated that human myogenic reserve cells (MuRC) generated in vitro are similar to Pax7+ muscle stem cells with a good regenerative potential in vivo. Recently, we showed that human MuRC are heterogenous for Pax7 expression with a Pax7High and Pax7Low subpopulation. At the present, we aim to characterize the metabolic and bioenergetic profile of human MuRC. We demonstrated that MuRC had lower metabolic activity as compared to proliferating myoblasts with an increase in fatty-acid oxidation and oxidative phosphorylation. Our results showed that the MuRC Pax7High subpopulation is less primed for myogenic differentiation with low metabolic needs. Altogether, we showed that MuRC Pax7High subpopulation is a promising source of cells for clinical trials. After finding a way to specifically isolate viable MuRC Pax7High subpopulation, we will further investigate their functional properties in vitro and in vivo after transplantation in immunodeficient dystrophic mice.
