Temporal response to a fast-refed paradigm within the arcuate nucleus and the median eminence

Abstract

To maintain body weight, energy intake (i.e., food intake) must, over time, equal energy expenditure: this process is known as energy balance. The role of the arcuate nucleus of the hypothalamus has been thoroughly studied and it has been proven to be a key regulator of food intake. Neurons but also glial cells have been shown to respond to circulating metabolic signals and to changes in food intake such as high-fat diets (Prevot et al, 2018; Langlet et al, 2013; Hsuchou et al, 2009). The aim of this project was to explore the interplay between these different actors in the regulation of food intake. We have implemented a fast-refed paradigm to analyse temporal changes in gene expression and cell morphology in adult mice. Our results show that there is indeed a temporal response in the ARH/ME region induced by fasting and refeeding and that glial cells, in particular tanycytes, may play a crucial role in response to refeeding. Morphological analysis of glial cells is needed to support these findings further.
