Characterization of human myogenic reserve cells

Abstract

Satellite cells (SC) are skeletal muscle stem cells essential for postnatal growth and regeneration of damaged muscle fibers. SC can be cultured and expanded in vitro, but their amplification, necessary for therapeutic uses, is associated with a reduction of their regenerative potential in vivo. Myogenic reserve cells (RC), generated in vitro, present numerous similarities with quiescent SC and are a promising cell source for muscle repair. In this master thesis, we tried to further characterise these RC, especially by studying their molecular signature and their metabolic status. For this purpose, we isolated human RC at different time of myogenic differentiation (48h to 96h) and we analysed them using flow cytometry, qRT-PCR and Western Blot. We showed that RC were quiescent cells locked in G0, capable to all proliferate when they were reactivated. At molecular level, we observed that RC upregulated NICD and EGFR, 2 important genes necessary to maintain stem cell quiescence. We also showed that RC adapted their metabolic status in vitro with a significant increase of AMPK, a key regulator in the energetic balance of the cell as well as an increase of autophagic flux. Taken together, human RC are quiescent cells locked in G0 similar to human SC, with a molecular and metabolic profile, which can explain, in part, their better capacities to survive and to regenerate in vivo.
