Organisms have evolved storing and signaling mechanisms to adapt to different environmental conditions. In plants, phosphorus is stored as inorganic phosphate (Pi) and as phytic acid or inositol hexakisphosphate (IP6). Pyrophosphosphorylation of IP6 gives rise to critical nutrient messengers named inositol pyrophosphates (PP-InsPs) which act as signaling molecules. PP-InsPs interact with proteins containing a SYG1/Pho81/XPR1 (SPX) domain. By interacting with these proteins, PP-InsPs can repress the transcription of Phosphate Starvation Response (PSR) genes and regulate the activity of proteins involved in Pi homeostasis. So far, only a few PP-InsPs interactors have been identified and characterized in plant model Arabidopsis thaliana. The aim of my master is to identify new components of PP-InsPs signaling in Arabidopsis. New candidates potentially interacting with PP-InsPs have been identified in a forward genetic screen and in a PP-InsPs interaction screen previously set-up in the lab which hosted me for the master thesis. Using the forward genetic screen, I identified a new genetic interaction between PP-InsPs biosynthetic enzymes, transcription factors regulating Pi starvation response, and a splicing factor named pre-mRNA Processing factor 4 Kinase A or PRP4KA. To confirm this genetic interaction, I generated knock-out mutations of PRP4KA using CRISPR-Cas9 genome editing technology in Arabidopsis. I further performed experiments to characterize the newly generated mutants and to provide preliminary data to understand the genetic interaction observed in the forward genetic screen. Overall, during my project, I studied a potential interaction between PP-InsPs and mRNA maturation and provided preliminary data and genetic material to further study PP-InsPs signaling in plants.
