The Triple Negative Breast Cancer (TNBC) represents only 15% of all breast cancer subtypes. Nevertheless, it is characterized by a high death rate. TNBC is highly aggressive and exhibits poor prognosis, being insensitive to conventional targeted anti-breast cancer treatments. It is therefore crucial to develop new therapies against TNBC. As in many other cancers, the signaling of the Wingless/Int (Wnt) family of growth factors is overly active in TNBC, being one of the causes of carcinogenesis. During my master a was a member of research laboratories located in the Department of Cell Physiology and Metabolism (PHYM) at the University of Geneva. One of them has hitherto identified a pool of compounds that target the Wnt pathway and has additionally shown very promising activities against TNBC cells in vitro and in vivo. To assess their toxicity, we decided to proceed by using high throughput screen assays on the Nematode C. elegans. The validation of the new protocol would allow to test the toxicity of a high number of molecules to employ in the preclinical development of the drug candidates against TNBC. My project is part of our laboratory pipeline of anti-Wnt anticancer drug discovery. Over the next few years, the compounds eventually emerging from this pipeline might become specific drugs, ultimately saving human lives from TNBC. Le cancer de seins impact fortement la longévité ainsi que la qualité de la vie de nombreuses femmes dans le monde. Bien que la variante connue sous le nom de Cancer du Sein Triple-Négatif (TNBC) constitue environ 15% de tous les cas de cancer de seins, son niveau de mortalité est dit proportionnellement élevé. Le TNBC est extrêmement agressif et se manifeste par des mauvais pronostiques, causant son insensibilité aux traitements conventionnels contre le cancer de seins de nos jours. Durant mon master, j'ai fait partie des laboratoires situés au département de physiologie cellulaire et métabolisme. Un d'entre eux a identifié un grand nombre de composants qui ciblent la voie de signalisation Wnt, ces composés montre une activité promettant contre les cellules TNBC in vitro et in vivo. Cependant afin d'approuver leur faible toxicité sur les cellules non-cancérogène je décide de procéder à un criblage à haut débit sur le Nématode C. elegans. La validation du nouveau protocole, permettrait de tester la toxicité d'un grand nombre de molécules tels que les nouveaux médicaments contre le TNBC. Ce projet fait partie de notre pipeline de laboratoire de découverte de médicaments anticancéreux anti-Wnt. Les composés émergeant dans les années à venir deviendront des médicaments qui sauveront de nombreuses vies humaines.
