Diplôme Master Establishing ChEC-Sort-Seq (CSS) to reveal protein binding dynamics during cell cycle in asynchronous cells eng 2020 author Cosandier Mathieu 805376 director Shore David M. 102587 The primary goal of this thesis is to present the use of three known techniques in molecular biology, ChEC (Chromatin Endogenous Cleavage), Sort (Sorting by FACS (Fluorescence Assisted Cell Sorting)) and Seq (High throughput Sequencing), and combine them to answer a technical as well as a biological question: Can these techniques be combined to reveal the binding dynamics of a protein of interest throughout the cell cycle? & Does Cdc6p interact in vivo with Rif1 and Mcm4 during the replication stage of the cell cycle? Originally, the present master thesis would have been an attempt to answer those two questions. Unfortunately, the vagaries of life sciences decided otherwise and thus the second inquiry changed as such: Analyzing Mcm4 and Orc4 binding dynamics in vivo to discover new potential origins of replication and unravel the temporal behavior of origin firing.
