Noncoding RNAs can in fact be activated in response to DNA damage. First we characterize the noncoding transcripts in the locus of the oncogene c-Myc and we tested their modulation after DNA damage induced by Trabectedin and Doxorubicin. We found antisense transcripts that were induced when we treated different cancer cell lines with DNA damaging drugs. The expression of c-Myc was consequently repressed. We found chromatin changes in the locus of the gene such as the phosphorylation of histone H2A.X and the displacement of histone H2A.Z, detected by chromatin immunoprecipitation experiments. Based on these data we speculated that the DNA damage-induced antisense RNA could be involved in the silencing of c-Myc in order to allow DNA damage repair.
