The most common is the 5’-m7 G cap, received by RNA polymerase II transcripts. However, if the RNA polymerase starts the transcription using a metabolite, like nicotinamide adenine dinucleotide (NAD), the RNA formed will have an alternative cap. The physiological roles and turnover of such NAD-capped RNAs are not well understood. It has been previously reported that human NUDT12 is a cytosolic enzyme capable of decapping m7 G-RNAs and NAD-capped RNAs, with a preference for the latter. The crystal structure of NUDT12 revealed that its homodimerization is essential for NUDT12 activity. It has also been reported that the cysteine protease bleomycin hydrolase (BLMH) is a direct interactor of NUDT12. Together, they form a 600 kDa dodecamer complex that is required for the localization of NUDT12 in cytoplasmic granules, distinct from P-bodies. My master thesis project aims to provide structural insights to the formation of this NUDT12-BLMH complex. Les transcrits d’ARN reçoivent une coiffe à leur extrémité 5’ afin de les protéger de la dégradation. La plus courante est la coiffe 5’-m7 G, ajoutée aux transcrits synthétisés par l’ARN polymérase II. Cependant, si l’ARN polymérase débute la transcription en utilisant un métabolite, comme le nicotinamide adénine dinucléotide (NAD), alors l’ARN ainsi formé portera une coiffe alternative. Les rôles physiologiques et la dégradation de ces ARNs coiffés de NAD restent encore mal compris. Il a été démontré précédemment que la protéine humaine NUDT12 est une enzyme cytosolique, capable de retirer les coiffes m7 G et NAD des ARNs, avec une préférence pour ces dernières. La structure cristalline de NUDT12 a révélé que son activité dépend de son homo-dimérisation. Il a également été montré que la protéase à cystéine nommée « bleomycin hydrolase » (BLMH) est un interacteur direct de NUDT12. Ensemble, ces deux protéines forment un complexe dodécamérique de 600 kDa, nécessaire à la localisation de NUDT12 dans des granules cytoplasmiques, distinctes des P-bodies. Mon projet de thèse de master vise à apporter des informations structurales sur la formation de ce complexe NUDT12-BLMH.
