This neuronal loss leads to the characteristic motor symptoms of PD, including tremors, bradykinesia, muscle rigidity, and postural instability. Although significant number of research has been made in understanding the genetic and environmental factors contributing to PD, the underlying cause of the disease remains largely elusive. Recent research has increasingly focused on identifying novel proteins and pathways involved in the disease’s progression, aiming to develop more e<ective therapeutic strategies. This study leverages the genetic tractability of Drosophila melanogaster to investigate the molecular mechanisms of PD, with a focus on the phagocytic Six-Microns- under (SIMU) protein, which emerged as a key factor in previous RNA sequencing analyses of parkin (park) null mutant, park1 flies, as a model of familial PD. Drosophila provides a powerful system for genetic studies due to its well-characterized genome, short life cycle, and conserved biological pathways relevant to human disease. Previous RNA sequencing data from park1 mutant flies identified a significant upregulation of SIMU, a protein that had not been previously associated with PD. These park1 mutants exhibit dopaminergic neuron degeneration and motor dysfunctions analogous to those observed in human PD patients. This study employs immunostaining and other molecular techniques to validate the upregulation of SIMU in park1 mutants and investigates its potential role in the neurodegenerative processes associated with PD. The findings suggest that SIMU absence may play a critical role in the mechanisms leading to neuronal death, possibly through pathways involved in neuronal maintenance or immune response. Understanding the function of SIMU in PD could open new avenues for therapeutic interventions, o<ering hope for slowing or halting the progression of this debilitating disease. This research contributes to the broader field of neurodegeneration by identifying and characterizing a novel factor involved in Parkinson's disease, providing a foundation for future studies to build upon.
