Implication of a putative small RNA in the regulation of OprD upon metal exposure and characterization of proteins involved in metal resistance in Pseudomonas aeruginosa

Abstract

In Pseudomonas aeruginosa, the OprD porin is responsible for imipenem resistance in presence of zinc. In this work we got interested in the Hfq-dependent regulation of the OprD and especially in an undescribed small RNA detected by RNA deep sequencing analyses that could be a potential partner of Hfq. So far, the experiments didn't allow us to describe a particular function for this small RNA in imipenem resistance or in metal resistance, however some hypotheses are discussed and remain to be verified. Comparing Cupriavidus metallidurans and P. aeruginosa's czc locus, we also tried to identify new proteins that would be implied in cadmium, zinc and cobalt resistance. Three genes were investigated: PA2807, PA0397 and an undescribed ORF (open reading frame). Our results confirm the implication of PA2807 in copper resistance, but not in cadmium, zinc or cobalt resistance. Nonetheless we showed that the three genes were induced in presence of zinc and this raises the question of co-regulation between resistances to different metals.
